The relationship between sisters is the theme of "Blood Memory" when Supergirl (Melissa Benoist) takes a trip with Nia (Nicole Maines) to her inclusive home town. After meeting Nia's sister (Hannah James), Kara discovers why her protege has been so hesitant to embrace the powers passed down from her mother (Kate Burton). At the same time the show begins exploring the aftereffects of Alex (Chyler Leigh) forgetting Supergirl's true identity from small memory gaps, to a lack of confidence, to being far less kind to the Maid of Might in the field. Bonding over their trouble with sisters, the episode works as both an origin story for Nia's eventual hero push while also staying true to the show's core principles of friendship and sisterhood as Kara reveals her secret to a friend whose turmoil she understands all too well.
While the episode does include some Agents of Liberty, "Blood Memory" also moves forward with the "other" Supergirl storyline making its first impact on the show when the Russian Supergirl somehow is the cause of super-powered some street drugs into monster steroids allowing everyone from frat boys to Agents of Liberty to take out their anger in the most destructive way possible. The other Supergirl's unexplained comatose state, and a call to the United States for help, also seems to be quickening her introduction into the main storyline. The other memorable plot point involves Superman's black friend James "Don't Call Me Jimmy" Olsen's (Mehcad Brooks) killing a cub reporter's story about Lena's (Katie McGrath) black-book genetic experiments leading James down a morally-ambiguous path that Supergirl likely would not support.
